Our Lufthansa flight from Newark to Prague gives us 45 minutes to change planes in Dusseldorf. Can we make the Prague flight?

If there is no delay then you have some chance.

As Ilja said, if no delays you should be fine. Last summer I made a connection with under 30 mins. Our inbound from Warsaw was late and then parked at a remote gate - which meant a bus to the terminal. We then had to go through passport control before rushing to the gate for our flight to Chicago. They were calling our names on the PA by the time we made it to the gate. We are in our seats with 5 mins to spare. My rush through the terminal makes me think it is quite compact and easy to get through. However, please do check if there are separate terminals for Schengen and non Schengen countries and if you will arrive in one and out the other.
