I am interested in going to Ephesus then to Pamukkale but am not sure how long that will take. I heard you have to stop in Denizli. I am looking for some kind of time table. I will be traveling in April. Any suggestions?

I wouldn't book via that website unless you have lots of money to burn. Busses in Turkey are really cheap--no need to pay those kind of prices!
Go to the Izmir otogar--this is the main bus station. If you are staying in Selcuk, you will need to take a dolmus minibus to the Izmir otogar. Then take your pick of busses to Pamukkale via Denizli. There are lots of them--you shouldn't have to wait more than an hour. Varan, Boss, Kamil Koc are three that run the route--there are certainly others. I particularly like Varan--the seats are comfy, there are two bus attendants to provide snacks and drinks, and some of the busses have Wifi!
You will have to change busses at Denizli, but that isn't really much of a problem.
