We will have one large/checked bag that we will need to store before and while on our Rick Steve's tour in June. The best place for us to store and retrieve that suitcase is at the Zürich HB. The sbb website shows a luggage storage location on Floor 0, but it can't be reserved ahead. Is there a chance that they would be full and unable to store our bag when we arrive?
Has anyone used this storage location? Experience? Recommendations?

Have you checked with the RS tour office to see whether you'd be able to stash your large bag in the hold of the bus and leave it there for the length of the tour? I haven't taken one of the tours, but I've seen references to stashing unneeded items in the hold of the bus. It would be at least trickier, and possibly impossible, if you don't have the same bus for the entire tour, of course.
I don't think the manned storage facility will be full, but you need to check the cost and the longest period that you can store your bags for. You can call them and I think they will find somebody who speaks English to answer your questions. Be prepared to provide measurements in metric.
Maximum locker rental time is 96 hours, so you would need a manned luggage storage.
There is a manned storage location at Zurich airport. They list rates by the week, and it is not cheap.

Where does the tour end?
You could just have SBB forward your luggage from the airport to where the tour ends, and have it waiting for you there.
