Would it be to drive to Granada and take a plane?
I'm confused. Why would you have to drive from Sevilla to Granada to fly to Lisbon. Couldn't you fly directly from Sevilla to Lisbon? It looks like TAP flies direct but you'd have to check the days and times to see if they're compatible with your schedule.
It looks like ryanair has a non-stop ( 3 or 4 days per week,depending on the month) and TAP has 2 daily non-stops. Seville to Lisbon
What I see between Granada and Lisbon is one stop flights.
I see no reason to drive to Granada to take a plane.

Take a flight from Sevilla to Lisbon. That’s what we did
An alternative to flying, is bus to either Tavira (a really nice town worth a couple days) and then train to Lisbon, or continue on the bus to Faro, and train to Lisbon. Probably 6-7 hours in total, but handy if you want a stop in the Algarve.
We flew TAP Air from Seville to Lisbon several years ago. Very easy. That's what I would do.
I guess you have to define “best.” Just a couple weeks ago I rented a car in Seville and drove to the Algarve area and spent a couple of nights in Lagos and enjoying the beaches and seafood before driving up to Lisbon and on to Porto.
