Four of us are traveling from Granada to Seville and from Seville to Madrid. Is it best to go by bus or train. How much longer is the trip one over the other

On the Seville-Madrid leg a train will be much, much, much faster. The train will take as little as 2 hr. 32 min. to cover the 320 miles. Obviously, a bus will take twice as long, or longer.
Rome2Rio will give you an idea of comparative travel times, but I've learned not to trust what it says about travel times, frequencies and fares. You really need to use Rome2Rio just as a starting point. If you keep drilling down through the web site you'll eventually find the name of the bus company(ies) serving the route, and you can get accurate schedule information from those websites. Train schedules should be checked on renfe.com.
There's a possibility of saving quite a bit of money on the rail tickets by buying early, but the cheapest tickets will be the most restricted, so you may not want to buy them super-early this year. Trains on the Madrid-Seville line do sometimes sell out, so it would be very risky (as well as probably costly) to wait to buy the ticket on the day of travel.
Between Seville and Madrid, the answer is obvious. The train is a lot faster.
From Granada to Seville it depends. The train is a bit faster and offers a bit more space. The bus on the other hand is a bit cheaper. My choice would be the train, but your priorities might be different.
