I plan to travel from either Madrid or the Basque County (Bilbao or San Sebastien) to Barcelona. (Madrid is the most likely departure city.) The date is June 12. Shall I pre-book tickets now?

Yes-as soon as possible if you’re interested in buying your ticket at a lower rather than higher price.
There are only a handful of high-speed trains from San Sebastián or Bilbao to Barcelona. and they sell out quickly. There are tons of high-speed trains between Madrid and Barcelona.
www.TheTrainline.com lists all train companies.
www.Renfe.com only lists Renfe trains.
