We're taking the Best of Spain tour in May. It ends in Seville and we will spend a couple of extra days there before flying out of Madrid. What is the best way to get to Madrid from Seville?
We plan on taking the train. Tickets usually open up 60 days before departure dates.
Direct trains run hourly between Sevilla S. Justa and Madrid P Atocha stations and take 2.5 hours. Discounts are available if you book tickets in advance through www.renfe.com (plan to pay with PayPal, to avoid credit card hassles) and outside agents like Rail Europe. There's no deadline to purchase, although occasionally possible for a train to fill up. Standard fare can go up to 77 euros per person in 2nd class, if lower rates sell out. That doesn't include another hourly (roughly) to transfer to the airport and of course check-in time once there, so most people would sleep in Madrid the last night before flying home.

I'd suggest taking the train. You may still be able to book discounted tickets on the Spanish Railways Renfe.com
I agree with Laura in terms of spending the night before your flight in Madrid.
Have a wonderful trip!
