Not the most scenic route for most of the drive, but the roads a good. I’ll assume you’re making the drive in one day.

Leave Toledo on the CM-42 and go to Consuegra. There isn’t much to the town, but it has a nice castle and windmills. From there, continue east on the CM-42, then get onto the CM-420 to Campo de Criptana. At Campo de Criptana are the windmills referenced in Cervantes’ Don Quixote.

From Campo de Criptana, if you had the time, continue east on the CM-420/N-420 to Belmonte Castle. It’s a very nicely restored castle and was used in parts of the 1960s movie El Cid. From there, work your way back to the E-5 and head south to Granada.

Depending on how leisurely you’d like to drive (meaning more than a day) you could stop in Almagro, Úbeda, or Jaén. Northwest of Granada is Alcalá la Real. It too has a castle.

There are plenty of options depending on how long you want to take getting to Granada.
