Whoa -- talk about a vague request? What do you want to do? A rest day or sightseeing? Where is your connecting flight? For us, a one day city visit would be Paris or maybe Rome.
Much of Spain is likely to be unpleasantly hot in the summer. I doubt that the cooler areas (the Basque Country, Galicia, etc.) offer stopover options for you. Paris may be out due to the Olympics this summer.
What, exactly, are the cities open to you as stopovers?
Yeah I knew it was a broad and random request. We don’t have flights booked yet. Thinking one city to visit w/ a major airport (besides Paris w/ Olympics) as a break to do some one day sight seeing. We’ve been to Italy, including Rome, and loved it.

If you stopped in Barcelona, the most popular site is probably Sagrada Familia. There are also Mediterranean beaches.
If you stopped in Madrid, the most famous site is probably the Prado art museum.
Both have good food and good wandering.
Consider a day in Athens, Greece to see the amazing Acropolis and New Acropolis Museum.
