Has anyone used onmytrip.com to travel from city to city in Eastern Europe? They offer door to door travel with an English speaking driver. The rate is a little less than traveling first class on the train. They also offer stops along the way. Sounds a little too good to be true!

No, I have not. But for me, a private transfer service is always a last resort - I prefer public transit especially trains to get from place to place, and a long train trip is actually a treat for me. Just a personal preference.
What kind of prices are you seeing for trips from where to where?
