Is it possible to travel by train from Coimbra to the Duoro Valley area, near Paso do Regua to spend a few days? The alternative is to go by train to Porto and rent a car and drive. Recommendations? Thanks
Very simple. Take the train to Porto then the train to Régua. The Douro valley trains run from the same station (Campanha) in Porto. Use cp.pt website to make reservations. There are a couple of coffee shops close to Campanha - in case you have to wait a bit between trains. Both trains run numerous times during the day.

Yes, there will be taxis but not many. If you are staying at a Quinta or hotel, they might be able to arrange a taxi to meet you at the station. It’s worth asking, especially if you are visiting during the peak summer season. The towns along the Douro also will have phone numbers for the taxi service in that area if you google it.
