I am flying into Lisbon and flying out of Porto 48 hours later. I would like to book a private guide from Lisbon to Porto with a few hours in Coimbra to see the university.

Try mydaytrip.com to hire a driver between the destinations, and you can add stops along the way. You could also train between these 3 destinations, of course.
