I booked a 6:40 am flight from Porto to London on a Sunday -- this is not until this summer but I'm wondering if it will be difficult to get an Uber or taxi so early on a Sunday morning? Or does the metro run that early?

Put Porto Portugal Airport Transportation in Google. There were a dozen Companies that you could book online.
Just needed your Porto address. I Have done this in Berlin and Rome and London and have had no problem getting an early morning cab to the airport.
They actually had an app for Berlin Cabs (talixo) and it updated me with how close the cab was and when they were at my apartment. It was slick.
We used Uber as we did everywhere in Porto. Most waits were 5-10 minutes.

I've also had to take an early flight out of Porto, and I'd recommend using a pre-arranged Taxi. The hotel staff made all the arrangements, and the driver was actually a bit early (despite the early hour). I've never used Uber so can't provide any comments on that.
I have booked a room in a near-by airport hotel the night before our flight, just to make it easy. There are several to choose from, some with-in walking distance to the airport.
Like Ken, I had my hotel arrange a taxi for an early AM flight. And it was at a discounted rate. At that hour on a Sunday, streets are empty and it should be a short, quick trip from the center to the airport.
No problem at all. Lot´s of uber, taxi, and so. Don´t worry. You even have metro after 6:00.
