Confusion re: testing and timing

If I am leaving America for Portugal on a Monday night but then have a lengthy layover in Amsterdam, do I calculate the timing of the PCR test before I leave America or before I board in Amsterdam? My flight to Amsterdam is not through TAP, but through United.
Leave Chicago 18:05 Monday
Arrive Amsterdam 9:05 Tuesday
Leave Amsterdam 17:20 Tuesday
Do I get the test 72 hours prior to my departure Monday or 72 hours prior to my departure Tuesday?

Thanks for your help. We don't leave until the first week in July, so if they drop the testing requirements, this post is moot. I just want to be ready and book the testing now.

dont over think. Get the test before you leave Chicago and it will be fine

When I log into the United site and visit their Travel Ready Center for my flight, it tells me the exact time that I need to get a PCR test and the exact time to get an antigen test - for my specific flights if they're all on United.
