Will be driving from San Sebastian area to the Douro valley. Any suggestions on an overnight between the two places? or even 2 nights if suggested.

León is a good midway stop on the fastest route, but it really depends on your pace. Do you want to stop along the way and spend some time exploring cities, towns, villages, and countryside? Do you want to drive along the coast? Or do you just want to limit your stops to the one or two nights along he way.
You could take a coastal route and spend a night in Oviedo and a second night in Santiago de Compostela. There are other routes you might consider, but they involve places I'm not familiar with, so I can't offer suggestions other than these.
I happen to be in Leon right now and it’s quite nice. We just arrived last evening so haven’t seen much, though. We drove from the Porto airport and it took about 4 hours.
If you want to go closer to the Douro to spend the night, either Guimarães or Braga are lovely.
León is nice with plenty to see to fill an entire day.
If you wanted to stay in a village closer to the valley rather than a city, Puebla de Sanabria is nice for a night. It has a small castle, and old church right on the main plaza and plenty of older buildings.
We stayed at the Paradors in each place.
