We will be on a NJ train from Ams to Offenburg, in a sleeper car, for the first time.
Offenburg is not the final stop.
Do I need to set a alarm to wake up before the predicted arrival time, as not to miss our stop ?

Are you in a sleeper, or in a couchette? In sleeper car the car attendant will ask when you want waken up, and when he should bring your breakfast. In the couchette I believe there is no breakfast included, so no wake up call.
But I would set an alarm nevertheless. I always do.

For an 0450 arrival I would definitely set an alarm clock, just in case!
