If I buy a ns train ticket from Schiphol to Delft,, single way, 2nd class, full fare, no subscription. online.
and take a Intercity Direct to Delft do I also need to buy a Intercity direct supplement ?

If you take the Intercity Direct, you also need to buy the supplement. However it doesn’t make much sense to take the Intercity Direct when you travel from Schiphol to Delft. The Intercity Direct requires you to change trains in Rotterdam. It’s much easier and faster to take the regular intercity from Schiphol to Delft.

as Dutch_traveler says. The high speed line trains bypass Delft so you would be backtracking and paying for the privilege.
The regular trains call Schiphol, Leiden, den Haag HS, Delft so much easier, cheaper and faster.
