Hello,
If I am making a trip to one city in NL to Arnhem and/or Otterlo, such as from Rotterdam, it looks like there will be multiple bus and train connections and transfers to complete. Do I need to pre-book the train or can I take my time and just miss connections and make them up?

The Netherlands has a nationwide mass transit network, with trains operating pretty much as subway does in the US (except better). And the to get to places the trains don't get to you take busses.
It is all wonderfully coordinated, and usually rather frequent. No need to pre book. Just tap in and out with your credit card (don't forget to tap out!) and use the "9292" app to plan your trip.
And yes, if you miss a connection you just take the next one.
