My wife and I will be there in late October for the first time. We'd like to get some kind of geographic/historical overview of the city. Ideally, a hop-on/hop off bus tour would be perfect. A driven tour of some sort with a guide would be perhaps a second option. My wife isn't able to walk for very long- we use wheelchairs for museums and such- so a walking tour is out of the question. Would a canal tour give us what we want? Is there such a thing?

Definitely take a canal tour. Ask your hotel’s concierge to arrange a driving tour.
Look up the CitySightseeing HOHO buses and boats.
We did two canal cruises on our trip in February. We did one of the usual ones through Flagship Amsterdam and were far less than impressed. Being an able bodied pedestrian it is difficult to navigate in Amsterdam; as you are constantly watching out for bicycles. Be on high alert if you attempt to walk anywhere with someone with limited mobility. I'm not sure how difficult navigating a wheelchair within those parameters will be, but always use extra caution.

Thanks for the recommendations, all. We have booked the canal tour with with Captain Jack, AND we are going to get a 48 hour pass for the hop-on/hop off boat and bus tour. BTW, thanks for the tips about the bicycles, too, we've heard a lot about them. And the wheelchair- we only use that for museum tours and such where she would have to be on her feet for an uncomfortably long time. She can walk about a bit, but we have to limit it both in terms of distance and speed.
If I remember correctly, the Anne Frank House tour involves some mildly steep
stairs up to the attic? Might be worth confirming things ahead because if you
want to book tickets, those go fast.
