My wife and I will be visiting Amsterdam for the first time in October and have reviewed the various segments of the city. Our goal is to enjoy the neighborhoods, renting some bikes, perhaps do a brief sightseeing cruise, see the van gogh museum and mostly soak in the joys of the city.
What neighborhood would you suggest we stay in that provides the best opportunity to enjoy the city? Also I'm looking for any suggestions for a moderately priced hotel as well as any sights worth seeing that I've overlooked.

I enjoyed the Hotel de Looier on the edge of the Jordan district. It is near the Anne Frank House which is worth a visit. There is a self guided walking tour from the Anne Frank House to the Resistance Museum which is worthwhile and informative, pick up a brochure with a map for €0.50 at Anne Frank.
I have been to Amsterdam perhaps 15 times since they late 1990's and I have stayed in a variety of hotels, The last 5 or 6 times there I have stayed at the Hotel Piet Hein. It is on a quiet street across from Vondel Park.. It is about a 5 minute walk from Leidseplein ( one of the major restaurant and bar districts) as well as less than a 10 minute walk from the Van Gogh and the Reijksmuseums.
I recommend you to visit the Jewish Historical Museum to get a taste of the Jewish life in Amsterdam. It includes the majestic seventeenth-century Portuguese Synagogue. For information: http://jck.nl/en/location/portuguese-synagogue
