Take the train to Delft and back. Forget my experience you should research the city to see if it interests you.

We just spent several days in Leiden, and loved it. Don't miss the windmill museum.

We also went to Den Haag to visit the Mauritshaus Museum and the M C Escher Museum. Well worth a side trip. Good lunch at the fish place in the train station there.

Both are easily accessible by train.

Do you have Rick Steves Amsterdam and the Netherlands? He describes a wide variety of day trips, from cities to small towns to rural areas, in far greater detail than anyone can in a forum post. He also includes goof-proof directions on how to see each place as a day trip from Amsterdam, and includes accommodations for some of them if you want to make it an overnight (particularly the longer daytrips like the Kröller-Müller Museum).

Personally, I liked Utrecht, Leiden, and Rotterdam, and was less taken with the Hague and Delft. I was interested in cities, though, so none of these places will appeal to you if you want more rural destinations.

Unless you go somewhere in the countryside, you'll get everywhere by using trains, which run with an amazing frequency. Familiarise yourself with the NS Railplanner to know which train to get and where you possibly have to change trains.
