Together with my mate, we are planning a bike trip. We are from Rotterdam and we and thought, that we go to Amsterdam. I searched the web for information on the distance and time our trips.

If you want to plan a complex bike route, and check all maps of bike routes, this is the website you want to check: http://routeplanner.fietsersbond.nl/ (English version is available though some info is badly translated)
There is no way to tell how long it would take to cycle from Rotterdam to Amsterdam. That greatly depends on your fitness, weather (wind), route chosen (it is forbidden to cycle alongside highways and controlled-access 2-lane roads, so you must pick bike routes not car routes) etc.
If you are really fit, you can cycle all the way in 4 hours, if wind is not a problem.
