We are flying into Rome and have booked a tour of the coliseum (arrive 9:00 am and tour at 12:00). Our tour ends at 3:00 and we plan on taking a train to Florence immediately afterward. My question is should we take the train from the airport and store our luggage there or take the time to get our luggage to Termini train station and leave from there? Is the train from the airport high speed?

There is only one direct train from FCO to Florence per day at 1:53
You’ll want to take the Leonardo Express to Termini, store your luggage there then consider a taxi to the Colosseum
Not sure you’ll actually make the tour on time
( next time don’t book important tour on arrival day)
After tour, return to Termini and take the next fast train to Florence ( Firenze SM Novella)

There are only limited trains directly from the airport to Florence, and all of those stop at Termini.
Basically, you will take the Leonardo Express train from the airport to Termini Station (The train only runs between the two).
Store your bags at Termini, then take the Metro, the Blue line, to Colosseo Stop, or walk, and find your tour. Do the same in reverse to Termini, or walk, retrieve your bags and find your train.
When searching for a ticket, your departure is Roma Termini, your arrival station is Firenze Santa Maria Novella or Firenze SMN.
There are many more trains leaving from Termini than the Airport, and no sense in backtracking. Buy your tickets directly from Trenitalia https://www.trenitalia.com/en.html , or you can check out their competitor Italo, https://www.italotreno.it/en Do not buy from third party sites.

I think you will need the travel angels on your side to make your tour on time.
If you do store your luggage, remember that there may be lineups to get yours out again.
Termini is exceedingly busy morning, noon and night.
Get back to the storage place well before your train leaves…….perhaps book your train to Florence no earlier than from about 5 pm?
When do plan on eating anything?
