Where to stop in Tuscany

Next month I will be touring italy with 3 other women. We actually rented a car and I've laid out our own itinerary. I've basically allowed an entire day to get from Rome to Florence and would like to stop one or two places along the way.

In planning, I've found a lot of wine tasting tours of the area, but I think they're generally designed as day trips from either Rome or Florence. I was envisioning that we'd drive ourselves to a winery and just take a tour when we got there (like we would in the US). Anyone know if this is an option? Better yet, anyone have any suggestions -- places you've been that would be good for us to stop at?

Wine seems like the most obvious opportunity to take a tour/tasting, but we're also interested in visiting a place where cheese or olive oil is made.

Take a map and follow the Route through the via Cassia (SR2) to Siena then the via Chiantigiana (SR222) to Florence. It's almost a 6 hour drive (excluding stops).
Do you also keep the car while in Florence? If so you need a hotel outside the ZTL.

We are, indeed, staying outside the ZTL in Florence.
Any suggestions about where we could stop to see how they make wine, cheese, olive oil, or similar?

Wine tastings...be aware that driving while under the influence is a BIG no no in Italy. Make sure you have a designated driver.

That out of the way here are some good stops. Starting in Montelpulcino then Montalcino travel up the 222
to Castellina in Chianti then to Greve in Chianti then on to Florence
