We are getting off a cruise ship in Genoa next April. Our group of 6 wants to spend a couple of nights here, but none of us have been there & wants some suggestion on where would be the best place to stay & what is a must see. Is 2 days enough time? We will be heading down the coast to Cinque Terre also.

You can stay in the historical center near the port. If you prefer to stay outside the city center, stay in the more residential upscale Nervi or even in Camogli, a suburb to the east. You can reach the city center by train from those locations.

There are many hotel and apartment options in the historic area. Keep in mind that the port of Genoa is huge and some cruise ships may dock a distance away from the historic area. If possible, find out in advance where your ship will dock and explore the transportation options to the "main" part of Genoa.
As far as "must see" sites, visit a library or bookstore and look through a few guide books that describe Genoa to find what appeals to you. Once the home for wealthy families involved in shipping, there are many old palaces as well as churches and a market to explore. Genoa is built on very steep hills and I suggest you try out the many lifts and funiculars that carry passengers up and down those hills.
I just want to say, make sure your hotel isn't REAL close to the port. I am told by my relatives who live in Genoa that it isn't safe there at night. But if you go in a ways and stay in the historic center, you will be fine. If you have a chance to go to the Albaro district it is our favorite area of Genoa. Walk along the promenade to a lovely cove area called Boccadasse. There are restaurants there and the best gelato in Italy!
the coast is nice - Albaro and Bocadasse. Not super touristy, but nice to walk around, plenty of places to eat and explore. easy to get to from the train, not far from the city. I would go back.
