The Vatican are will be more convenient because it is much closer to everything than Travestere. Look at a map. Travestere is quieter, more residential, better restaurants, etc. and would be my preference of the two.

Generally I would pick Trastevere (not "Travestere") over the Vatican area, but this particular B&B in Trastevere is at the very far south end of that neighborhood, so it's well away from the major sites. Why are these your only two options?

We have been inquiring about places that can fit 4 adults comfortably. These two seemed like some of the nicer places for a good price. There have been other options that we are going through. Do you have any suggestions for the best region of Rome to stay in for a 2 day stay?
My husbands parents are 70 and 64.

If it's your first visit to Rome, then the historic center is what most people would recommend. The area that includes Campo de' Fiori, Piazza Navona, and the Pantheon. What's your price range in dollars or euros, and when are you going to be there? Any mobility issues?

The outstanding "Ron in Rome" has excellent information on exactly this sort of question. Read all about it here on his website. The piece is called "The "Best" Place to Stay in Rome is..." Happy travels.
