Next summer we will have 4 days in Venice and then rent a car and take two days to drive to Pienza in southern Tuscany. I know it's possible to drive it in half a day but we want to see a new part of Italy. What would you suggest seeing along the way and where would you suggest staying? It doesn't have to be the most direct route. Maybe Ravenna? Assisi? Or if not a city, any special B&B along the way?

Assisi is a great stop for sure. Stay in town even tho parking maybe a little difficult. the true magic of Assisi comes out at night.
You will travel thru Umbrian hills to get here . However a quickstop in Ravenna to see the mosaics is also a gd visit.

Ravenna if you like beautiful mosaics and a nice city center, and Assisi. Close third choice would be Todi, but it may be too similar to Pienza.
