Will be leaving the CT on a Tuesday morning, heading to Castellina in Chianti, and then leaving Castellina in Chianti for Venice on Thursday morning. What's the best location to pick up and drop off the car? I was thinking we'd take a train from CT to La Spezia, Pisa, or Florence, pick up a car in one of those towns, and drive the rest of the way to Chianti. Then maybe drop off the car Thursday morning in Florence and take a train the rest of the way to Venice? Thoughts? Thanks in advance for your advice!

I'd get the car in La Spezia. It's adjacent to CT and it's only a 2-hour drive from there to Castellina in Chianti. No point in going to Pisa unless you want to see Pisa - which you could certainly do by car anyway. From Castellina to Venice is only a 3 1/2 hour drive - I'd be tempted to just keep the car and drop it off in Venice. To me that'd be easier than having to return a car in Florence and then catch a train. Both drives would be mostly on divided highways and not difficult at all.
