Hi All,
I'll be traveling by bus in Tuscany and have read that not all bus drivers sell tickets on-board–true? Some stops I'll be using have no ticket counter or Tabacchi nearby. Anyone have experience with this? In particular, I need to get from (a stop near) San Gimignano through Poggibonsi to Siena. Cheers!

Yes, you should be able to.
I traveled this route in reverse (round trip) Siena to San Gimignano, and bought my tickets at the bus station in Siena.
