I will be spending a few nights (4 nights each) at Venice, Florence, and Rome. When would be the best time to buy train tickets for each journey? Should I hold off on buying them until I get to each city, and visit the train station the day before I leave? Or should I buy them all before I leave the US?
You will receive different opinions. If you are planning on taking high speed trains and know when you'd like to travel, I always buy in advance to take advantage of savings if they are available. These tickets can not be changed or refunded so I only do this when I am sure that I am going at that particular time. When possible I prefer to take the high speed trains but to visit the smaller towns (you have only listed the big cities) I take the regional trains and buy them a day in advance online to make my travel easier. There is no savings on the regional tickets.

Oh the day, up to 15 minutes before the scheduled departure time, if you want maximum flexibility.
Going to the station the day before is the worst option: Most likely all Discounted tickets will be sold out, so You'd waste vacation time to pay the same fare you'd pay on the day of travel. You might as well get tickets at a travel agency paying their service fee, at least you'd save the time to go to the station.
Note that wherever you get tickets, you can get all of them at once. FCO station included.

Any idea when trips after June 11 will be uploaded? We need a high speed train Venice to Florence June 13th.
June 11 is the date this year the new train schedules for dates thereafter will be uploaded and sold. You may see them on Trenitalia’s website 30 days prior to that date.
