I have 3 full days in Puglia region early Sept 2024.
Flying into Brindisi and renting a car.
Before I pull out all the guide books and watch all the RS I can handle, I need expert opinions on where to stay for 3 nights for 2 people on a 250usd/night budget, where to dine, explore, eat and drink like a local and see the most beautiful things.

I don’t believe that RS includes Puglia in any of his guidebooks or videos. The guidebooks you may have to pull out could include Lonely Planet, Rough Guide, Fodor’s, and Frommers.
I’ve not made it to Puglia, yet, so I’ll be doing likewise. Have good trip!
We spent ten wonderful days in Polignano a Mare at Hotel Covo dei Saraceni .Give this town a look along with Lecce.I am happy to hear you are renting a car for exploring Puglia.
We ate at seafood restaurants as much as we could.
A very good one was Miravigloso, also both Antiche Mura and Casa Mia were also very good.
You will hear the song. “ Volare” being sung as Domenico Modigno was born and raised there and his statue attracts lots of visitors.

Short answer: Ostuni old town, very close to Brindisi, great restaurants
