We are scheduled for the Scavi tour in Oct. I read on their website that you cannot bring in large bags or purses. I usually carry a 16 x 12 leather tote bag/purse would they consider that too large?

It's probably OK. Backpacks are absolutely not allowed, but I got in with a shoulder bag not much smaller than yours.
