Visiting Italy for the 1st time

My wife and I are visiting Italy for 3.5 days in early June (flying in/out of Milan). We are mostly interested in the ambience of Italy -- sitting at sidewalk cafes, strolling through quaint streets and shops, eating classic Italian food etc. Medium interest in history/art/museums. We're trying to pick between Venice, Florence/Tuscany, Cinque Terre. Any suggestions? Thanks!

If the choice is from among the 3 you list, Florence/Tuscany would be my suggestion.

Any would be fine.

Had you considered Lake Como? Given your short stay, its close with easy access from Milan

For only 3.5 days maybe think about Cinque Terra and Bellagio on Lake Como for a beautiful relaxing stay with delicious food and amazing scenery...
Or if you want to see history and museums then you could spend all you time in Florence...

Stresa or Bergamo are within easy reach of Milano.

Como popped into my mind to considering the length of your stay and that you're in and out of Milan. Out of your list Florence/Tuscany would definitely be my choice. Florence can be seen in a fairly short time, you just might be pushing it if you try to cram in much else since you have to get to and from Milan and might want to sleep too. Florence has a lot to offer.

I'm with Zoe. Venice, Florence and CT are too far away. The ambience of Italy is everywhere, you don't have to go to the usual suspects to encounter it.

Voting with Zoe and DougMac here too: Bergamo, Stresa or even Varenna would be within easy reach of Mlian for a trip as short as the one you'll be taking. Venice, Florence and the CT are too far away and should be saved for a future trip when you have more time and/or can fly more directly into those regions.
