Aside from the bus could one take a taxi from Rome. And I would it be hard to find one to return to Rome. A few years ago four of us used a car service, it was very pleasant but cost 300 euros. Looking for a less expensive alternative beside the bus. Or is the bus a fun way to go.
Train to Milan, then train to city of Como, driver from Como. Or driver from Milan.
Or fly Rome to Milan LIN, driver to Villa d’Est.

I would take the train and walk to the villa.
Have taken the bus to Villa d’Este 2 times. It was easy. Had to get to the bus stop in Rome the board the bus and get off by the Villa. Don’t remember the exact details but should be easy to google. The bus is the only reasonable choice unless you have$$ or time to burn. Pick a sunny day to enjoy the fountains. The Villa and gardens are spectacular. Think the Italian version of French chateau.
I don't understand how young professionals can afford to take an Uber home from a bar every night of their lives. The bus and the train are both fine ways to get to Villa d'Este. You could say the bus is better for Hadrian's Villa. Our bus (summer) was jammed, and we stood most of the way. You could also say it's a drag to take the Metro to Ponte Mammolo.
This newsboard is mostly budget travelers. I would never take a car service to Villa d'Este from Rome. I will admit that we took (Rick's recommended .... !) car service from Prague to Cesky Krumlov, but we needed to do it in one day, with as much time there as possible. That is much farther than Tivoli from Rome.

The train is easy (from Termini or Tiburtina) and then when you arrive in Tivoli follow the signs to Villa d'este. It's maybe 10-15 minute easy walk through the town and sign posted all the way.
I remember it was easy and cheap, and to confirm I've just checked and the trains run around every half hour and cost 3 euro. It's a small station, so simple to navigate the return trip too.
