I’m travelling from Vernazza to Nice this September. I am struggling finding the best (easiest) train route. I’ve heard there may be a direct train from Genoa to Nice but nothing comes up when i investigate on Trainline. I am travelling on a Sunday if that makes any difference.

It is too soon to look up train times for September, but the typical route is Vernazza-Sestri Levante -Genoa (Principe)-Ventimiglia-Nice. There is no other train route, just sometimes you could connect somewhere else between Vernazza and Genoa.
Yes, I took the direct train from Genoa to Nice in 2017. You may be able to get a direct train from Vernazza to Genoa, but it may be faster to take the local train from Vernazza to Monterosso or Levanto first and connect there to an IC train. Not all trains stop in little Vernazza.
Check the schedule for next week. If you see the trains you would want to take, most likely you'll be able to take the same trains in September once the schedules are updated for September.
On Sep 3, there’s a train that departs Vernazza at 6:19a that requires two changes arriving in Nice at 12:11p: https://www.trenitalia.com/en.html.
Apparently the Thello direct trains don't run between Genoa and Nice anymore...don't see them on the schedule even for this week.
