Train. If you are intending to go straight to Rome, bus from Marco Polo airport to Mestre station, and then train to Rome.
Mestre is the closest station to Venice, but is on the mainland. Mestre works OK if you are not wanting to travel into Venice.
There is no need to go to actual Venice (Venezia Santa Lucia station) to get a Rome train.
Mestre to Roma Centrale is about four hours on a very comfortable train.
Check out the Italo trains. https://www.italotreno.it/en. They are really nice, and if you buy your ticket far enough in advance, even first class is a deal.
If you don't want to see Venice you could take a flight from Venice to Rome, and then the train into the centre.
