Venice to Florence or Venice to Tuscany and back to Florence

We are planning on traveling from Venice to Tuscany. However, we have two options. Would it be easier to take the train to Florence, spend a full day there (2 nights), and then rent a car and head south towards Sienna in the countryside? Or, drive straight to the Sienna area from Venice and visit Florence as a day trip?

Staying in Florence for 2 nights would require us to stay 3 nights instead of 4 in Tuscany near Sienna, and 1 less night somewhere else later in the trip. Not Staying in Florence would require an hour of travel each way to visit the city as a day trip, but at least we wouldn't have to move hotels.

We were planning on the one brutal day itinerary of Florence since we are more into hiking/scenery/food, but can't miss visiting the city for the highlights.

Since you'll have a car if going straight to Siena area, I think visiting Florence as a day trip would be the better option. With the car, you can arrive as early as you want and leave as late as you want, allowing for a long day of touring. It saves you a change of hotel, with the downside being that the driver should skip wine if having dinner in Florence. In most cases I'd suggest staying in Florence, but your particulars lead me to suggest the opposite.
