We will be picking up our Avis hire car from Piazzale Roma - just wondering what would be the best way to get there with suitcases in tow from our accommodation near the Rialto -there will be 5 of us. Just wondering if anyone would know if we can catch a vaporette and just walk to this location.

You can walk to the nearest vaporetto stop and take the #1 or #2 vaporetto to P.le Roma. Then from P.le Roma you would walk to the car hire office. It should not be very far off the main area. Current cost is 6.50 euro per person and that includes luggage.
You could also take a private water taxi to P.le Roma, but you'd still have to walk to the office. Cost would probably be in the 50-to 60 euro range.
