After spending two days in Venice, we will be picking up a rental car and driving to Siena. What is a good location (city, airport?) to pick up a rental? Don't mind taking the train or taxi to a location.

The closest place would the "Venice Downtown" location at Piazzale Roma. To see these options on AutoEurope.com, it looks like you have to select Venice All Locations first, then choose the pick up spot in a later step.

Just in case you haven't seen the warnings in other threads, people have noted that while picking up at Piazzale Roma is easy, there are speed cameras on the causeway connecting it to the mainland. You will get a ticket if you are even a little bit over the limit, as well as an administrative fee from your rental company for turning your information over to the ticketing agency. (Your driving speed is being monitoring in lots of places in Italy, by several different methods, but apparently the cameras on that causeway are particularly unforgiving). You take a vaporetto right to Piazzale Roma, so it's much easier getting a car there than elsewhere.
