Trains don't serve Pienza, so the most efficient way is probably train to Siena (via Florence), then bus to Pienza. The provincial bus service, Train S.p.a (yes, an odd name in English) offers several buses per day. Click here to see their schedules in pdf format. Scroll down to route 112/A for outbound trips from Siena, and 112/R for trips back to Siena from Pienza. Note there are several different stops in Siena itself. One-way fare should be about 5 or 6 Euro.

Check a train to Chiusi then a bus to Montepuliciano then to Pienza.
