Why Mestre? It's usually less expensive than Venice, but not very charming. You could stay in Padova instead (about an hour from Venice, less than 10 euro each way by train), or stay in a monastery in Venice (monasterystays.com, about 70 euro per night). If you stay in Venice, buy the vaporetto pass (travel in Venice is expensive, about 7 euro per ride) for about 35 euro for 3 days, less for one or two days - a bargain if you plan to daytrip out of Venice. The trip to Murano/Burano involves at least 3 tickets if you buy them separately, whether you stay in Venice or in Mestre. Florence, La Spezia/Cinque Terre is way too much for one day, in my opinion. If you do stay in Venice rather than Mestre, the earliest train may leave before the first vaporetto run.
You can check exact train fares on the Trenitalia site, which gives different price options for trains. Regionale trains may not be listed on the website, but they are usually cheaper although they make more stops.

Hmmm... Pretty ambitous (and exhausting) schedule. Leave out La Spezia and CT and, if you depart very early, you just might be able to get a photo of the leaning tower and a few hours in Florence and be home before midnight. Really, only 3 days in Venice and you want to spend one day riding the rails? IMHO 3 days is only scratching the surface in Venice.

It is 5 to 7 hours each way from Mestre to the Cinque Terre. If you leave Mestre at 6:32 AM, you arrive La Spezia at 13:05. You still have to take a local train to the CT town of your choice. It's another 5-7 hours going back. Stop in Pisa for an hour to see the tower? OK. But Florence for an hour is a joke. What are you going to see in an hour? So 12 hours on a train to make what, three one hour stops? If you want to see Florence go to Florence and spend the night (or 2 or 3 nights). If you want to see the CT, do the same. While you are in Venice experience all that Venice has to offer! It's a wonderful, magical place! I do agree a day seeing Murano, Burano and Torcello is worthwhile. Be sure your buy a vaporetto pass. Oh, and the cost of your desired trip to Florence, Pisa and the CT is €35-60 depending on trains and available discounts. That's a ONE WAY fare.

I don't really think the Florence, Cinque Terre, Pisa day can be done. We did The Cinque Terre as a day trip from Florence and that was a pretty long day. But if you need convincing, the easiest thing to do is to go to http://www.italiarail.com/ and look up your connections. Leave at 5:44 am arrive Pisa 9:03. Leave Pisa at 11:00 and arrive Florence at noon. Leave Florence late in the afternoon or early evening and return by bedtime. Doesn't sound like much fun to me, but it might be done. Add Cinque Terre and you have a very long train ride and nothing much to show for it.

I don't often say this on the webiste but a 1-day visit to Florence, Pisa and the CT from Venice is just not reasonable to consider. First, it's a 5.5hr train journey each way to and from the CT. The fastest runs will go through Florence so you will get to see Florence from inside the train station. However, if you eliminate the CT, you really can do a 1-day trip to see the leaning tower and some parts of Florence. Here's my suggestion. Wait until you get to Venice to buy your train tickets. Your train journey will involve two train tickets - a high-speed train from Venice to Florence with seat reservations and a Regional train from Florence to Pisa with no seat reservations. This will be very easy to do. The train journey time to Pisa is just 3hrs19mins. Here's a LINK to the morning train schedule for Aug 24. It will be much same for any of the 3 days you are in Venice. At Venezia Mestre, get on the high-speed to Florence but don't validate your second ticket yet. Do that when you get to Florence and you are changing trains. The second ticket is for the Regionale train. It's good for 60 days until you validate it. Once validated, its only good for 6 hours. You can see the fares on the link. At Pisa Centrale, it's a 25-minute walk to the tower and 25-minutes back. You can expect to spend 2.0hrs-2.5hrs to see the tower and take pictures. If you take the early train, it's about 13:00 when you get back to the train station. Buy a ticket for the Regionale train back to Florence. 7.10E. Trip will take 1hr15min. Your in Florence by 14:30. You now have time to spend 4-5 hours in Florence. Go back to the train station and buy your high-speed ticket back to Venice. Last high-speed 21:25.

trying to understand your one letter words is hard work but if I get the gist, yes you can attempt to sleep on the train and leave Mestre at midnight:21, change at Arezzo 4 hours later (11 minutes to change) and arrive in Firenze at 2 minutes before 6 am. You could also leave at 3:26, direct, arriving at 6:15. They both arrive before 7:40. They would get you to Pisa (note there is only one "s") before 8am. You have been looking at the Trenitalia website for yourself, right? They have all the connections and all the prices.

You will not likely get discounted tickets at the station in Italy. They will most likely be gone as they are limited. You will pay the base fare. If you go online now and look at the fares, you will see the same base fare. No differences in the base fare. However, looking early online, you will also see the discounted fares if they are available. You can see these in the link that I posted above. Again, you won't see those discounted fares at the station.
