I hope I am understanding this correctly, I will be landing at the Venice airport. If I purchase a 72 hour pass on veniceconnected.com I will be able to take the ACTV Bus to the St Lucia train station then hop onto the vaporetto all with the same pass-yes???

Yes, the pass covers the orange ACTV bus from the airport as well as the vaporetto. It goes to Piazzale Roma where you can catch a vaporetto. The train station is on the other side of the canal.

Yes, it is fairly well marked. Plus most of the people on the bus will be going there as well.
Basically P.le Roma is a large bus parking lot. After you exit the bus, turn your back to Venice and face the causeway you came in on. The vaporetto stop is on your right towards the front of the lot.

One more question, will I be able to pick up the #1 to the Rialto from this stop or will I need to transfer?

Yes, no transfer needed. The vaporetto will be travelling from your left to your right as you face the canal. So the front of the boat will be to your right as you board.
