Hello, we are arriving Venice in early September, visiting Lake Garda and Dolomites for 10 days, then need transportation from the airport car rental to the San Basilio terminal to catch our ferry to Rovinj. Should we take an Alilaguna boat then a vaporetto? We really don't want to walk our luggage 10km across the island.

I think a little easier would be to take the No 5 bus to Piazzale Roma, then either the No 2 or No 8 boat to San Basilio. You can get a combined Bus+Boat ticket for this. The bus leaves from the front of the airport, then then a short walk to the Piazzale Roma vaporetto dock. The Alilaguna dock is a ways from the airport, although a moving walkway is in the skybridge.
The Bus+Boat ticket is 18 EUR pp, same price as an Alilaguna ticket, which drops you at Zattere, then either a long walk to San Basilio, or a 9.50 EUR vaporetto ride.
Thanks, bus may be easier. Can we just jump aboard and pay with credit card or should we look for an ATVO kiosk at the airport? And then we may just walk to the ferry terminal, it's 1.2km with a few great restaurants along the way.
No, you need a ticket. There is a kiosk at the bus stop in front of the airport entrance, or buy a ticket at the desk inside. Ticket is 10 EUR pp.
Walking from Piazzale Roma to San Basilio will involve many stepped bridges. Hauling luggage, it would not be my choice, but doable.
