I have an early flight out of Venice. 6:30am. Would it make the most sense to stay one night near St Mark’s square and one night near Piazzale Roma, take either bus or a taxi to the airport.
From Piazzale Roma it's faster.
Stay in just 1 place
Near P Roma makes the most sense
Or stay 1 night in Venice, 1 night near airport

The airport doesn’t open until 4am. When you arrive in Venice, while still at the airport, go to your carrier’s help desk and ask them at what time you should arrive for your return flight. Last early flight I had was 7am in Turin. Arrived at airport at 5:30. The agents didn’t show at the ticket counters till 6:30.
You'll make life a lot easier for yourself by staying at an airport hotel the night before. Your Venice hotel should hold your luggage for you after check out, until you pick it up in the evening.
Rather than throw away a night at an airport hotel, I suggest you choose a hotel near Piazzale Roma for both nights. There are some nice options there, actually; some right on a little canal with plenty of Venetian charm.
I am not a fan of staying “in” San Marco because of the crowds. For sightseeing there, you can easily take the vaporetto from Piazzale Roma (the #1 and #2 start here so you generally board an almost-empty boat, unlike the scrum you face at later vaporetto stops). Or have a little adventure finding your way there on foot,through the back streets of Santa Croce and San Polo.
We are staying at 3* Hotel Arlecchino for one night on our next trip; right next door is Hotel Olimpia with 4 stars. Previously we have stayed at the AC Hotel Venezia, a Marriott property, but it is very expensive. In the morning you can walk to Piazzale Roma for an early bus to the airport, or ask your hotel to call a taxi (which will cost around 50€).
Took Lola’s advice. I booked hotel for 2 nights near Piazzale Roma. This will allow me to maximize my time sightseeing in Venice and not worry about getting to airport.
