I've researched hotels in Verona, Padua, and Mantua as a base from which to visit those cities and Vicenza (and wherever we feel like going!) Can someone recommend which would be preferable?
It depends on whether you'll have a car or rely on public transit. I like Vicenza because it's very accesible by car and in the midst of the Veneto.

Yes, it depends on your mode of transportation. I have made the swing from Venice to Milan, stopping in Padua, Vicenza, and Verona. All are fine places to stay. Vicenza is the least touristed and Verona the most. Padua is worth a stay as it is a college town and has a slightly different feel. If you have a car, Verona is a headache to park near the old city center (which is where you will want to stay). Padua is a bit easier, but Vicenza might be the best with your own wheels. Check out hotel prices and inquire about parking options. Italy is always a headache for drivers. If you can pack light I recommend the train and any of the cities makes a viable base. Bravo for hitting Mantova (Mantua) as it is un-touristed by non-Italians. However, if I recall correctly it is not as accessible to all of the other cities you mention. When I visited, I made it a day trip on my way out of Verona towards Bologna. I hope this helps. Happy travels.
