When you get your tickets in advance, which is advised, you will be able to pick your date and entry time. Keep in mind that Vatican Museum tickets go on sale 60 days out from your desired date.

If you do not buy your tickets in advance, you will only have higher-cost options (like commercial tours) available to you, or a long wait in line that might not even result in a ticket. Even in the winter, online tickets for the Vatican Museums sell out in advance. At busier times of the year, they can sell out weeks in advance. I think I've seen the least expensive, basic-entry tickets sold out for more than a month.
