The N route is specifically there to run around the main island all night long. Therefore, there is vaporetto service 24 hours a day. The question didn't ask if specific routes run 24 hours, just if there is vaporetto service through the night.
Umm..Where's the "hike"? The Linea N does an efficient route around the main island of Venice including through the Grand Canal and even goes to the Giuedecca and the Lido. Where's the "hike"?
It simply runs less frequently than service during the day. But we've established that - in response to the original poster's question - that Venice has vaporetto service for 24 hours a day.
Wow this is a pretty funny thread :) Here is a better question ~ what time should we plan on heading back to our hotel in general? Midnight?

Venice isn't all that big. You may find that walking back to the hotel is no biggie.
Out of curiosity, what do you expect to keep you out in Venice until that late at night?
Keep in mind that if there is a rail strike vaporettos also go on strike. We encountered this a few years ago. Very interesting getting to our hotel.
Chani makes a valid point Alex. Venice does not have much in the way of late night hot spots. Walkways and campos start to get deserted around 10 pm.
The city is fun and fascinating to wander around in late and in the wee hours but you won't find a lot of places that cater to late night revelry.
We were in Venice in early June and it was so quiet on our way home from dinner at 10:00-10:30 at night, that we felt compelled to whisper in the streets.
Venice is not a late-night party spot, so don't worry too much about getting home after midnight. A good street map for walking is much more valuable than a vaporetto schedule in my experience.
Along with a good map, a compass is a real asset in Venice. Day or night, it is pretty hard to keep your bearings once you are out of sight of the Grand Canal.
The only time I almost missed the "last" vaporetto was during Carnivale.
thanks everyone ~ just remembering our last trip where we didn't have dinner until 8 or 9, then strolled around (which I hear is great to do in Venice at night), got gelato etc so was just wondering if vaporetto were still running at 10-11pm time. Sounds like they are!
