There is so much to see in a day! I would suggest driving through the Val d'Orcia. Get off the autostrada and take the backroads. Where are you starting from? If you are beginning around Siena then drive south on the R2. You will get a much better feel for the countryside. Most interesting mid-sized towns are Pienza (greatly underrated by Rick), Montalcino and Montepulciano. If you only have a day, you should probably just pick one to explore. Pienza is fun to walk through and see the shops. If you go there, be sure to finish your visit by walking along the back side of the town that overlooks the Val d'Orcia - follow the signs that say "passeggiata." In addition to one of these small cities, try to drive through (and get out and walk around of course) a small hill town like Montisi. That's plenty for a day. Italy is not meant to be rushed!
