Any suggestions for agriturismos if we do not have a car. We are planning to fly into Rome then spend a few days in Cinque Terre. From there we are open but would love to explore Tuscany (maybe Lucca and Siena). Any thoughts or suggestions would be amazing.
Agriturismos require a car.
Most agriturismos assume a car as your transportation.
There are a limited few "nearish" to towns such as those that advertise "a 30-45 minute walk to Siena" but that's hardly practical. You would definitely need to take a cab or shuttle there with luggage and then depend on walking/biking into town to reach a bus or a train for transportation anywhere else. If your goal is to stay primary at the agriturismo and you might want to take a trip into the city that might be fine. But if you want to use an agriturismo as a base to explore you'll need a car or at least a reliable shuttle to someplace with mass transit.
The other solution would be arrange for someone to take you from your agriturismo around the countryside as a tour guide. As long as you can get to and from your place the beginning and end of your trip you could replace a car for exploring with having someone else do the driving. I'm sure some places offer this - or at least a list of recommendations - as a service.
Rural Tuscany is not well served by mass transit which is part of its charm but makes visiting as a tourist problematic.

As mentioned, a car would be best for rural Tuscany. That said, you could still have an amazing stay at The Isabella Experience just outside of Castelmuzio. We stayed there with my family of four (husband and two kids) + my parents this past summer. Isabella is very accommodating and may have a driver she can connect you with.

Otherwise, Siena would be a wonderful stay. I haven't been to Luca but have heard great things and I don't think you need a car to get there.
As above, it would be easier for you to explore some of the region without a car if you base in a town/city with good rail connections. Florence usually tops the list of bases for day trips around Tuscany as it's a central transport hub. Lucca can be reached in abt. 90 minutes by rail; Siena is about the same. You can also take a "rapid" bus to Siena that lets you off closer to the center than the train.
Other easy day trips are possible as well, depending on time and interest.
Wonderful suggestions. The Isabella looks amazing! Any thoughts on ability to rent a car in Lucca, Siena or La Spezia? At this time, we do not plan to go to Florence as we have already been and are trying to explore new places. Thank you for all of your help and insights!

Yes there are car rental agencies in all those locations although hours will be shorter and availability may be limited (automatic for example)
