After spending a few days in Florence, we plan to explore the best of Tuscany by car for an additional 6 days. We would like to include a visit and perhaps overnight in Assisi. In or near which towns do you suggest we stay if we want to divide these 6 nights between two locations?
We did 2 nights Siena, 2 Montepulciano then 2 in Assisi
Dropped car in Spoleto and took train to Rome
Worked well
We picked up car at Sansovino Hertz location (outside ZTLs) thru AutoEurope

The IDP is required to be licensed to drive. Car rented at Via Palagio degli Spini, the complex near the Florence airport, reached from town by taxi. Maybe in Chianti first, then further down, around Montepulciano and Pienza:

You could dump a Europcar rental in Foligno; train to Assisi 20 minutes. From the Assisi station, Bus C up to town; tickets from the station shop. There are no decent rental agencies in Assisi.
You say Tuscany but are planning to go
to Umbria where Assisi is. So , to clarify, you now also want a base in Tuscany? Consider Panzano, Siena, Pienza,Cortona.
In Tuscany, there is Volterra, San G, Lucca to the north. Assisi is in Umbria, but you could also go to Perugia, Spello, Montalcino, and Orvieto.
